Porcine reproductive and respiratory syndrome virus (PRRSV) is an arterial RNA virus first identified in the United States in 1987. PRRSV virions consist of a positive-strand RNA genome and a nucleocapsid protein (N). The genome of PRRSV is an unsegmented single-stranded positive-stranded RNA of approximately 15 kb in length with a 5′ end cap structure and a 3′ end Poly(A) tail. The nucleocapsid protein envelope of PRRSV contains six structural proteins: membrane protein M, small envelope protein E, N-glycosylated glycoproteins GP 2, GP 3, GP 4 and GP 5. Porcine reproductive and respiratory syndrome (PRRS) is a highly contagious infectious disease caused by PRRSV, characterized by reproductive disorders in pregnant sows and respiratory symptoms in piglets, which seriously affects pig populations around the world and causes considerable economic losses to the pig industry.

Effective surveillance of PRRSV infection requires sample collection in the field and accurate testing in the laboratory. Several serological methods have been developed to detect PRRSV-specific antibodies in pig serum to determine PRRSV infection status in herds and individual pigs, such as: indirect immunofluorescence assay (IFA), immunoperoxidase monolayer assay (IPMA), and enzyme-linked immunosorbent assay (ELISA). IPMA and IFA require replication of the virus and are labor intensive, are not suitable for routine testing on a large scale. In contrast, ELISA is simple, rapid, sensitive and specific, is the gold standard for the diagnosis of porcine PRRSV infection in the pig industry.
