Avian Astrovirus PCR Kit Development

Astrovirus (AstV) is an unenveloped, single-stranded RNA circular virus that was first identified in 1975 in a stool sample from a child with diarrhea. Since its first discovery, astrovirus has been isolated from a variety of mammalian and avian species. Currently, six different astroviruses have been isolated in birds, are TAstV-1, TAstV-2, ANV, CAstV, DAstV-1 and DAstV-2, based on the species of origin and viral genome characteristics. AstV can infect a variety of poultry and cause outbreaks of intestinal diseases, posing a serious threat to poultry health and causing huge economic losses to the poultry industry. Therefore, it is necessary to develop a rapid and accurate method for detecting AstV.

Avian Astrovirus PCR Kit Development

Avian astroviruses can cause related diseases after infecting most animals, such as poultry enteritis death syndrome, broiler kidney pain, broiler stunting syndrome and fatal hepatitis in ducklings. The detection of astrovirus in most species is limited to electron microscopy (EM) and fluorescent antibody detection. However, the sensitivity of EM is low, and the virus can be detected only in the peak period. Fluorescent antibody test may not be able to identify avian astroviruses with different serotypes. PCR significantly increases the sensitivity of detection compared to EM, fluorescent antibody assays, and enables researchers to develop specific avian astroviruses detection kits. PCR has become a very common tool for diagnosing avian astroviruses infection.

Workflow of RT-PCR Kit Development

Workflow Details
Viral RNA extraction Extract the viral RNA from the avian astrovirus positive sample.
RNA-polyacrylamide gel electrophoresis (RNA-PAGE) The extracted RNA of avian astrovirus was subjected to RNA-PAGE.
Preparation of complementary DNA Synthesis cDNA by reverse transcription from avian astrovirus RNA for primer designing.
Designing of AstV specific primers Primer of avian astrovirus can be designed using professional software and database.
Optimization of RT-PCR assay We use a partial plasmid of the specific AstV gene as a template for optimizing and evaluating the sensitivity of the PCR assay.
Assessment of PCR efficiency Independently validate the AstV PCR assay by interlaboratory comparison tests.
