Neospora caninum ELISA Kit Development

Neospora caninum is a parasitic protozoan belonging to the class Sporozoa and the genus Neospora, parasitic on cattle, sheep, dogs and other animals. Canines (domestic dogs, mountain dogs, wolves, grey wolves and wild dogs) are the ultimate hosts of N. caninum. Neosporosis is a disease caused by N. caninum that is widespread worldwide and primarily causes abortion or stillbirth in pregnant animals as well as motility disorders and neurological disorders in newborns. N. caninum is a major cause of abortion in livestock worldwide and causes serious economic losses to the farming industry. Therefore, the establishment of an effective method to detect the infection of N. caninum plays an important role in the prevention and control of neosporosis.

Neospora caninum ELISA Kit Development

Currently, a variety of serological methods have been developed for the study of N. caninum in animals, including direct agglutination, indirect fluorescent antibody test (IFAT), Western blot and enzyme-linked immunosorbent assay (ELISA). ELISA is a commonly used method for serological detection, which detects N. caninum by specific binding of antigens to antibodies to be tested in serum. This method has the advantages of simple operation, short reaction time and high detection throughput, and is widely used in large-scale screening of animal neosporosis.

Workflow of ELISA Kit Development

Workflow Details
Submit ELISA kit development requests Determine the ELISA development protocol and estimate the cost and cycle based on the assay targets and experimental requirements.
Antigen preparation Design and synthesize primers for the nucleic acid sequence encoding the antigenic protein and obtain the target fragment by amplification, connect the target fragment to the expression vector, induce expression to obtain the recombinant protein, and carry out SDS-PAGE and Western blot to identify its immunogenicity, purification recombinant protein.
Antibody preparation Preparation of antibodies specific for dense granule antigen and stage-specific proteins of N. caninum, analysis for antibodies specificity and epitope.
ELISA kit development Optimizing the final reaction conditions, evaluation of sensitivity, specificity and concordance of ELISA kit.
