Caprine Arthritis Encephalitis Virus ELISA Kit Development

Caprine arthritis encephalitis virus (CAEV) is an enveloped, single-stranded positive-stranded RNA virus that belongs to the genus Lentiviridae in the family Retroviridae. The genome structure of CAEV is complex and can encode four structural proteins, core protein P28, P19, P16 and envelope glycoprotein gP135. CAEV mainly infects goats and is the causative agent of goat arthritic encephalitis (CAE). CAE is a chronic inflammatory disease affecting the lungs, joints, udders, and central nervous system of small ruminants such as sheep and goats. It is widely distributed around the world, with serologically positive detection rates of up to 65%-77% in the United States, Switzerland, Canada, Australia and France. The widespread spread of CAE has had a serious impact on the health of goats and brought significant economic losses to the goat breeding industry.

Caprine Arthritis Encephalitis Virus ELISA Kit Development

Currently, early laboratory detection is used to identify CAEV-infected animals and then to take measures for CAEV prevention and control. Several tests have been used for CAEV detection, among which agar gel immunodiffusion (AGID) and enzyme-linked immunosorbent assay (ELISA) are recommended by the World Organization for Animal Health (OIE). AGID is suitable for early screening of CAEV, but the number of antibodies is low in the early stages of infection and may not be detected resulting in false negative results. In addition, the AGID test cannot detect CAEV infection in milk samples. In contrast, ELISA has been shown to be more sensitive than AGID and is the most commonly used method for laboratory diagnosis of CAEV infection in most countries.

Workflow of ELISA Kit Development

Workflow Details
Submit ELISA kit development requests Determine the ELISA development protocol and estimate the cost and cycle based on the assay targets and experimental requirements.
Antigen preparation Recombinant E. coli cells bearing P16 (or P19, P28, gp135)gene were cultured in liquid medium and induced to express the antigen. Then the antigen is purified and the purity and immunogenicity of P16 (or P19, P28, gp135) were examined by SDS-PAGE and Western blot.
Antibody preparation Preparation of monoclonal antibodies specific for individual antigens of CAEV or antibodies against whole virus multi-antigens.
ELISA kit development ELISA procedure, optimization of ELISA working conditions, validation of ELISA kits.
ELISAkit delivery We will provide you with customized ELISA kits and product quality inspection report.
