Newcastle disease virus (NDV) is a single-stranded, non-segmented RNA virus belonging to the family Paramyxoviridae. The NDV genome contains six major genes, which can encode six structural and two nonstructural proteins. The host range of NDV is wide. It is reported that 27 of 50 species of birds can be infected with NDV. Newcastle disease (ND) is a poultry disease caused by NDV with high mortality and morbidity causing considerable economic losses to the world poultry industry.

Currently, a variety of serological assays have been developed for NDV diagnosis. The hemagglutination inhibition (HI) is considered the standard laboratory test for Newcastle disease and is the most widely used serological method for measuring anti-NDV antibodies in poultry serum. However, when serum samples are contaminated, higher false-positive results are generated and the sensitivity of the method is lower. Enzyme-linked immunosorbent assay (ELISA) has the advantages of easy standardization, high-throughput detection, high sensitivity and no need for serum pretreatment. It is an effective tool for NDV detection and can meet the requirements of epidemic prevention.
