Ruminant Abortion AGID Kit Development

Abortion is the artificial termination of pregnancy after organogenesis is complete but before the fetus is viable. Reproductive failure due to abortion disease is a significant revenue drain in many ruminant livestock production systems. Recurrent abortion is a common disease in ruminants. However, in most situations, an abortion rate higher than 5% to 8% is usually unacceptable and worthy of investigation. The commonly diagnosed infectious causes of abortion in ruminant animals are Coxiella burnetii, Chlamydia abortus, Brucella spp., Leptospira spp., Campylobacter fetus, Listeria spp. and Toxoplasma gondii. Some viruses can also cause abortion such as bovine herpesvirus type 1 (BHV-1) and bovine herpesvirus type 4 (BHV-4). The management and control of ruminant abortion outbreaks is important in limiting their spread, and in preventing zoonotic infections.

Ruminant Abortion AGID Kit Development

Given that rapid and accurate laboratory diagnosis is central to controlling abortion outbreaks, the submission of tissue samples to laboratories offering the most appropriate tests is essential. Direct antigen and/or DNA detection methods are the currently preferred methods of reaching an aetiological diagnosis. Among them, agar gel immunodiffusion assay is a commonly used technique for the detection of pathogens that cause ruminant abortion.

AGID test detects antibodies produced in response to infection, and it is compatible with samples from serum and plasma. Several AGID methods have been developed for the detection of pathogens (bacterial & viral) that cause abortion in ruminants, including single radial immunodiffusion test and double immunodiffusion test.

Workflow of Ruminant Abortion AGID Kit Development

Workflow Details
Serum sample collection Serum samples were obtained from ruminants and left to clot at a specific temperature.
Gel preparation Agarose gel was prepared in a specific buffered saline solution at a specific temperature for a given period.
Preparation of agar plates Dispense the melted agar onto a flat surface and prepare the agar plate at a specific temperature.
Agar gel immunodiffusion for antibody detection For each batch of specimens, a strong positive, weak positive, and negative reference serum will be included to aid in interpreting results.
Interpretation of test results for antibody detection Interpret the result via different analysis methods and evaluate the efficiency of the assay.
