Clostridium perfringens PCR Kit Development

Clostridium perfringens is a Gram-positive anaerobic rod-shaped bacterium that widely exists in the natural environment and in the intestinal tract of humans and animals. It is an important pathogen of humans and various animals, which can cause necrotizing enteritis, enterotoxemia and other diseases. In recent years, the world's major poultry countries and regions are widespread necrotic enteritis (NE) caused by Clostridium perfringens, especially in Europe and the United States and other developed countries, with the ban on antibiotic growth promoters, the disease is becoming increasingly serious. Clostridium perfringens not only reduces animal production performance and increases chicken mortality, but also causes human infection through poultry meat products, resulting in huge economic losses and human food safety issues. Therefore, the development of a rapid and accurate detection method for Clostridium perfringens is crucial for the control and treatment of NE.

Clostridium perfringens PCR Kit Development

In recent years, with the development of molecular diagnostic technology, a variety of molecular methods have been used to characterize bacteria, including temperature gradient gel electrophoresis (TGGE), terminal restriction fragment length polymorphism (T-RFLP), denaturing gradient gel electrophoresis Gel electrophoresis (DGGE) and conventional PCR. However, these methods cannot perform quantitative analysis of Clostridium perfringens, which limits their application. In contrast, real-time PCR was used to enumerate Clostridium perfringens in the gastrointestinal tract and feces of animals due to its ability to quantify bacteria. In addition, researchers have developed a variety of PCR methods based on the different detection needs of Clostridium perfringens, including RT-PCR, multiplex PCR, qRT-PCR, IMS-PCR, etc.

Workflow of Clostridium perfringens PCR Kit Development

Workflow Details
RNA extraction Purify the virus and extract RNA from Clostridium perfringens-infected cell cultures.
Primer design A set of primers can be designed according to the sequences of specific strains of Clostridium perfringens using professional software.
Establishment of PCR conditions Maintenance of thermocycling conditions to conduct amplification and detection through PCR.
Reproducibility evaluation of the PCR assay The reproducibility of the PCR kit was assessed by determining the intra-assay and inter-assay coefficients of variation (CV).
Assessment of sensitivity and specificity The specificity of PCR kit was assessed by detection of RNA from Clostridium perfringens. The sensitivity of the PCR kit was evaluated by determining the limit of detection (LoD).
