Pigs typically suffer from viral gastrointestinal infections such as porcine coronaviral enteritis and porcine epidemic diarrhea (PED). The duodenum, jejunum, ileum and colon's epithelial cells are where the PED virus (PEDV) mostly infects and replicates. Additional assays are needed for effective diagnosis and therapy as acute manifestations can not differentiate between several swine gastrointestinal virus infections. Using fibroblast as targeted cells, the virus neutralization assay inhibits the viral attachment to the cell and reverse transcription and integration. To find group-specific antibodies against gastrointestinal disorders in pigs, the virus neutralization (VN) test has been widely used. The virus neutralization methods have the advantage of being able to accurately evaluate serum samples that have been highly hemolyzed and cytotoxic. In recent years, many antibodies have been found using VN technologies such as plaque reduction neutralization test (PRNT) and serum virus neutralization assay (SVN).

Workflow of Porcine Gastrointestinal Virus VN Kit Development

Workflow Details
Virus titration Working dilution of the stock virus was determined and the stock virus was aliquoted in multiple tubes and stored at a specific temperature. 
Dilution The stock virus was diluted in a virus diluent with trypsin and titration was performed in quadruplicate.
Incubation Incubation was performed at a specific temperature at a given time with a limited supply of carbon dioxide. 
Cell-fixation Media was removed from virus titration.
Micro-neutralization assay All serum was treated with receptor destroying enzyme before use in assay and stored at a specific range of temperature for a long term.
