Salmonella pullorum Plate Agglutination Test Kit Development

Salmonella pullorum is a small gram-negative bacillus, usually single, occasionally two or more together. The bacteria do not form capsules and spores, and have no flagella and no motility. Pullorum disease (PD) is a global poultry disease caused by Salmonella pullorum infection. It has caused huge economic losses worldwide due to its high mortality and decreased egg production. Although countries are adopting various strategies to control Salmonella pullorum infection, persistent multi-drug resistance and novel Salmonella mutation make it still a serious threat to the poultry industry.

Salmonella pullorum Plate Agglutination Test Kit Development

The diagnosis of pullorum disease usually requires the isolation and identification of Salmonella pullorum. At present, the detection and diagnosis of this disease are still mainly done by traditional bacteriological methods, but the many steps of bacterial culture and isolation, and the complicated types of biochemical reactions make the detection process of Salmonella pullorum complicated, time-consuming and laborious, which is increasingly unable to meet the needs of the rapidly developing trade of poultry and their products. In order to seek a simpler, faster and more accurate detection method, researchers have established a variety of detection methods such as serology, immunology and molecular biology on the basis of traditional Salmonella pullorum detection methods. Serological methods such as plate agglutination tests are widely used for the detection of Salmonella pullorum because of their cheapness, operability and rapidity.

Workflow of Salmonella pullorum Plate Agglutination Test Kit Development

Workflow Details
Sample collection Samples were collected from animals infected with Salmonella pullorum.
Production of seeds Samples were inoculated on Mycoplasma agar, and after a period of incubation at 37°C, a sample from each single colony was transferred to Mycoplasma broth and incubated at 37°C.
Preparation of chromogenic antigens Neotetrazoles and tetrazolium staining antigens were prepared for use, and the concentration of staining antigens was measured by the Folin Phenol method.
