Mycoplasma synoviae (MS) is a polymorphic spherical organism, smaller than Mycoplasma gallisepticum (MG). There is only one serotype of MS, and the pathogenicity of different strains varies. The symptoms caused also vary depending on the tropism of the pathogen. MS is one of the important pathogens infecting chickens and turkeys, which can cause acute or chronic infectious synovitis or airsacculitis. When it is mixed with Escherichia coli, Newcastle disease virus, infectious bronchitis virus, etc., it can lead to more serious diseases. MS infection of poultry causes poor growth, increased mortality and a dramatic drop in egg production. This causes serious economic losses to the poultry farming industry. Therefore, the development of a rapid and effective method for detecting MS is of great importance for the prevention and control of poultry diseases.

Plate agglutination test is a serological test in which agglutination occurs after the binding of particulate antigen and corresponding antibody. Under the action of electrolytes, antigen and antibody complexes, after a certain period of time, form clots visible to the naked eye. PAT is often used in the diagnosis of MS because of its simplicity and rapidity.

Workflow | Details |
---|---|
Sample collection | Samples were collected from animals infected with Mycoplasma synoviae. |
Production of seeds | Samples were inoculated on Mycoplasma agar, and after a period of incubation at 37°C, a sample from each single colony was transferred to Mycoplasma broth and incubated at 37°C. |
Preparation of chromogenic antigens | Neotetrazoles and tetrazolium staining antigens were prepared for use, and the concentration of staining antigens was measured by the Folin Phenol method. |
Mycoplasma synoviae PAT kit development | Development of Mycoplasma synoviae plate agglutination test kit using staining antigen. |
