Immunoglobulins are heterodimeric proteins composed of two heavy chains (H) and two light chains (L), belonging to the eponymous immunoglobulin super-family (IgSF). By analyzing the amino acid sequences of the heavy and light chains of different immunoglobulins, it was found that the composition and arrangement of amino acid residues near the amino terminal (N-terminal) of the heavy and light chains vary greatly, and this region is called the variable region (V region), while the composition and arrangement of amino acid residues near the carboxyl terminal (C terminal) are relatively stable, so it is called the constant region (C region). Each V or C domain consists of about 110-130 amino acids, averaging 12,000-13,000 kDa. Each heavy and light chain of the immunoglobulin molecule can form a spherical domain by intra-chain disulfide bonds to perform its corresponding function, so it is also called the functional region. The heavy chain and light chain V region each form one structural domain, which is denoted as VH and VL respectively. The heavy chain and light chain C region form several structural domains with different numbers as follows: 1) γ, α and δ heavy chains form three structural domains, CH1, CH2 and CH3; 2) μ and ε heavy chains form four structural domains, CH1, CH2, CH3 and CH4; 3) the light chain C region has only one structural domain, which is denoted as CL.
